Adhesion Behavior of Chondrocyte and Fibroblast on Specific Peptide-immobilized PLGA Film

Abstract
Poly (lactic-co-glycolic acid) (PLGA) has been widely used in tissue engineering because of its biocompatibility and biodegradation. However, its hydrophobicity typically has some disadventages in the cell adhesion, proliferation, and differentiation. Therefore, this study aims the surface modification of PLGA film using peptide grafting technology. To achieve this purpose, plasma treatment technique was used and then RGD and GVMGFO peptides were chemically grafted through the reactions between carboxylic acid and amine group. For the cell adhesion on these surface-modified PLGA, chondrocytes and fibroblasts were seeded on PLGA-PAA-RGD and PLGA-PAA-GVMGFO films, respectively and then cultured for 24 hours in incubators. The ratio of chondrocyte adhesion increased in the order of the adhesion, PLGA£¼PLGA-PAA£¼PLGA-PAA-GVMGFO. Meanwhile, the number of fibroblast adhered to PLGA-PAA-RGD films was greater than that on the PLGA-PAA-GVMGFO films. Therefore, the peptide grafting on PLGA might be suitable to improve the cell adhesion for tissue regeneration. (J Korean Wound Management Soc 2009;5:88-93)
